黃秋霞與林國榮是一對真心相愛，並已珠胎暗結，可惜他們的良緣遭到林母的反對，一對薄命鴛鴦被無情拆散。林國榮在母親的壓力下，另娶妻子娟娟，但娟娟經醫檢確定不能生育。為了延續林家香火，林母軟硬兼施，要回了秋霞含辛茹苦養育的兒子志強。幼小的志強到林家後，因思念母親偷跑回家，被風吹雨淋，高燒不退昏迷不醒。秋霞誤以為志強已死，傷心過度神經錯亂，被送進了療養院。十八年後，志強來到了療養院工作，意外地見到了生母，當他唱起幼時的兒歌，秋霞竟奇跡般的恢復了正常，母子終於團聚。（這是在網上找的一段介紹，不知是記憶失真還是怎麼回事，印象中好像不是這樣的。）
母親被迫與相依為命的兒子分離，變成了瘋子。這部80年代放映的台灣電影讓觀眾們哭濕了一塊塊手絹，母子分離那刻， 「世上只有媽媽好」的歌聲響起，銀幕上下一片號啕聲。據說此片讓觀眾第一次有了進電影院帶手絹的准備。
就故事內容而言，《媽媽再愛我一次》是台、港通俗影視產品中最典型的家庭倫理大悲劇，在50和60年代，類似劇情的影片不斷被搬演，也的確曾經感動過不少觀眾。但近20年，這類影片已被視為「老套」而很少有人拍攝，縱使偶有這類作品推出，也是反應冷淡收場。事實上，就在此片在台灣推出公映的同一年，台灣另一家製片公司也拍攝了由陳榮樹導演的同類型影片《媽媽帶我回家》，結果兩部影片同樣得不到台灣觀眾的重視，映了幾天便靜悄悄下片了，在海外市場更連公映的機會都沒有。不料失之東隅收之桑榆，兩年之後，《媽媽再愛我一次》渡海揚威，竟在大陸上以黑馬姿態造成空前轟動。
